Mark Lee is well-known for his performances in several popular television series and films in Singapore. He gained fame for portraying Lee Tok Kong in the Mediacorp Channel 5 sitcom Police & Thief, and made guest appearances as Lee Kok Peng in the comedy series Phua Chu Kang Pte Ltd from seasons five to seven. Additionally, he played Su Hao in the long-running drama Holland V on Mediacorp Channel 8. Mark also received acclaim for collaborating with Jack Neo and Henry Thia in films like Money No Enough and Liang Po Po: The Movie, as well as in the enduring comedy variety show Comedy Nite (搞笑行动). His directorial skills were showcased in the comedy horror feature The Ghosts Must Be Crazy.

In 2010, alongside his longtime collaborator Christopher Lee, Mark was honored with the prestigious All Time Favourite Artiste award at the Star Awards, marking him as the first host-comedian to receive this accolade, as previous recipients were mainly actors. Consequently, he is no longer eligible for the Top 10 Most Popular Male Artistes awards in future ceremonies.

While Mark initially gained recognition for his comedic roles, he has expanded his career to include hosting various programs, predominantly in infotainment, travel, and DIY. He co-hosted the well-liked home makeover show Home Decor Survivor and the infotainment series Behind Every Job with Bryan Wong, which garnered multiple nominations in hosting categories at the Star Awards. In 2012, he secured his fourth Best Variety Host award for his popular series It's a Small World (season 2).

Typically, Mark has been given the screen names Ong or Huang in Jack Neo's films, with exceptions for Liang Po Po: The Movie, That One Not Enough, and Ah Long Pte Ltd.
